Ranolazine: No apparent proarrhythmic characteristics

  • There appears to be no evidence that ranolazine has any effect on either the trigger or substrate of torsade de pointes.

  • Ranolazine was not shown to cause EADs over concentration range 1-300 µM (wide range of models) and it suppressed EADs induced by proarrhythmic agents such as sotalol (eg, studies in Purkinje fibers, cardiomyocytes, whole rabbit hearts).

  • Ranolazine was found to have no effect on QT dispersion in humans at 500-1500 mg bid (MARISA trial).
