Ranolazine: Long-term use
- Studies on ranolazine safety and tolerability longer than 3 months are lacking.
- Koren et al investigated whether age, sex, diabetes, history of MI and HF, and exercise duration were predictors of discontinuation due to adverse events in a longer term (≥2 years) study of high risk, angina-limited patients.
- Mortality rates (2.8% per patient year) compared favorably to those predicted by the Duke Treadmill Score (5.25%).
- The majority of patients tolerated ranolazine therapy for at least 2 years.
- Age >64 years and history of HF, but not male sex, diabetes, history of MI, or below median exercise tolerance, were significant predictors of drug intolerance.
