Evaluation of fasudil in stable angina: Trial design
- Fasudil is approved in Japan for prevention of cerebral vasospasm following subarachnoid hemorrhage. It has not been approved in the US.
- Recently, Vicari et al reported on a trial of fasudil as adjunctive antianginal therapy in US patients. This was a dose-escalation study, with fasudil force-titrated from 20 mg tid to 80 mg tid in 20-mg increments. Nitroglycerin as needed and a beta-blocker or calcium channel blocker were allowed.
- Mean age was 64.4 years (placebo) and 66 years (fasudil). The proportion of men was 74% and 76%, respectively.
- Symptom-limited exercise treadmill testing was performed at peak and trough after each dosing period.
- The primary outcome was the change in exercise test duration.
